MtHsp70 interacts stably with mitochondrial Hsp60 and Hsp10. WT and mtHsp70_His yeast cells were grown in the presence or absence of cycloheximide (CHX) as described under “Experimental Procedures.” Mitochondria (Mito) were isolated and subjected to affinity purification via Ni²⁺-NTA-agarose. Bound proteins were eluted with imidazole and analyzed by SDS-PAGE, Western blotting, and immunodetection with the indicated antisera. Load, 0.5%; elution, 100%.
