TFP inhibits cell migration without affecting protease activity. (A) Transwell migration assays reveal inhibition of cancer cell migration among PC3 cells and C4-2b by TFP at 10 μM concentration. (B and C) Treatment of HT1080 cells with TFP results in a significant decrease in cell migration based on wound healing analysis, while exhibiting no effect by TFP on NIH/3T3 wound closure. NIS-Elements imaging software was used for wound closure analysis. Representative images are shown. (D) TFP has no effect on HT1080 cell proteolytic activities as examined by a fluorescent substrate degradation assay. (E) TFP treatment of HT1080 cells does not influence the expression of MMP-2 and MMP-9 and does not influence the activation of either of the gelatinases as observed in gelatin zymography.
