WT BALB/c and IL-13 KO mice (n = 4–5/ group) were prime-boost immunized with i.n. FPV-HIV/ i.m. VV-HIV. Fourteen days post booster vaccination mRNA expression profiles in gut and splenic KdGag197–205-specific CD8+ T cells were evaluated using Fluidigm 48.48 dynamic arrays as described in methods. WT BALB/c spleen (SP) and Peyer’s patch (PP) n = 6 pools of hundred KdGag197–205-specific CD8+ T cells are indicated by blue circle and triangle respectively. IL-13 KO mice spleen n = 5 and Peyer’s patch n = 9 pools of hundred KdGag197–205-specific CD8+ T cells are indicated by red circles and triangles respectively. Data indicate log mRNA expression of IFN-γ (a) and TNF-α (b) normalised to housekeeping gene L32. The experiment was repeated three times. Data represent the three experiments.
