EPCR-dependent biomarker regulation in cultured cells. (A) GM-CSF–elicited BMDCs from wild-type or EPCR-deficient mice were treated for 3 hours with LPS (100 ng/mL) and mRNA levels of Irf8, Peli1, Ccl22, and Malt1 relative to 18S rRNA were measured by quantitative PCR. Data are expressed as the fold-difference relative to untreated wild-type cells and represent average ± standard deviation from 4 independent culture experiments. (B) RAW cells were treated for 3 hours with the indicated TLR ligands, and the abundance of Peli1 mRNA relative to 18S rRNA was measured by RT-PCR. Data represent the average ± standard deviation from 2 independent experiments with triplicate measurements of each sample. *P < .01 by pairwise comparison with levels in the untreated sample (Student 2-tailed t test).
