A. Ly294002 blocks the effects of TGF-β1 on SERT function. Caco-2 monolayers were pre-treated with Ly294002 (50 μM) for 30 min and then with TGF-β1 (10 ng/ml) for another 1h in the presence or absence of inhibitor. N = 3. * P<0.01 vs control. Immunoblotting was performed utilizing phospho-specific P85 antibody. The blots were stripped and re-probed with the total anti-P85 antibody to normalize for equal loading of protein in each lane.
