EI-03 regulates cytokine production by DCs. (A-H) Bone-marrow derived DCs were stimulated with Mtb (50 μg/ml) for 6 hours with the indicated concentrations of EI-03 or the DMSO control. Cells were lyzed for RNA extraction and cultural supernatants were collected for protein quantification. Relative mRNA levels of TNFα (A), IL-6 (B), IL-10 (C) and IL-1β (D) were measured by real-time PCR. Protein levels of each cytokine (E-H) were quantitated by ELISA (*, p < 0.05; **, p < 0.01 as compared to the DMSO control). For measuring expression of co-stimulatory molecules and MHCII expression, Mtb-stimulated DCs with or without EI-03 were analyzed for CD80 (I), CD86 (J) and MHC II (K) levels with flow cytometry with respective antibodies. The results shown are representative of three independent experiments.
