The gametocyte culture was purified using a Nycoprep cushion, and stage V gametocytes (8,000 per well) were plated onto 384-well plates into which compounds had been predispensed. After 24 to 48 h of incubation at 37°C, the gametocytes were activated, followed by 24 h of incubation at 26°C with antibody against Pfs25 labeled with Cy3. Activated female gametocytes were detected by the Cy3 fluorescence signal in the Opera high-content screening system.
