Potentiation of macrophage phagocytosis of tumor cells by SIRPα blockade. A schematic of SIRPα blockade by soluble high affinity CD47 is shown. Left, CD47 expression on cancer cells activates SIRPα on macrophages, recruiting SHP-1 and -2 tyrosine phosphatases and preventing macrophage phagocytosis of cancer cells. Right, soluble high affinity CD47 prevents CD47 on the cancer cell surface from engaging SIRPα on macrophages, thereby acting as a competitive antagonist.
