(A) The Adhfn6 allele of Alcohol dehydrogenase contains a splice site mutation which retains an in-frame stop codon in the intron. This causes the Adhfn6 mRNA to undergo NMD, during which the 5′ fragment is degraded by the exosome and the 3′ fragment is degraded by Pacman. (B) To estimate the level of Pacman function in the pcm5 and pcm14 alleles, the level of the fragment degraded by Pacman was compared between lines containing the Adhfn6 allele and either pcm5 or pcm14 (n≥11. p<0.001 for all comparisons unless indicated, ns = not significant.
