Tyrphostin AG1296 inhibits cell migration of PLX4032-resistant cells.

Notes: (A) A375R cells were treated with tyrphostin AG1296 (0.0625, 0.25 or 1 μM) for 8 hours. The non-migrated cells on the upper surface of the filter were removed, and the migrated cells on the lower side were stained and photographed. The representative images are shown. Cells were then lysed and colorimetric determination was done at 595 nm. (B) Quantitation of the inhibition from Transwell assay. (C) A scratch was introduced into a monolayer of A375R cells, followed by treatment with tyrphostin AG1296 (0.0625, 0.25, or 1 μM) for 8 hours. The width of the wounded cell monolayer was measured in five random fields, and representative images are shown. (D) Quantitation of inhibition from wound healing assay.
