TNF-α inhibits BMP-2-induced mesenchymal C2C12 cell differentiation to osteoblasts. (A) ALP activity assay. C2C12 cells treated with 200 ng/ml BMP-2 revealed an increase in ALP activity; however, in cells treated with a combination of 200 ng/ml BMP-2 and 10 ng/ml TNF-α, the elevation of ALP activity was inhibited. (B–D) Gene expression assays for Col I, Ocn and Bsp indicated that the BMP-2-induced increase in gene expression was inhibited by 10 ng/ml TNF-α. **P<0.01, ***P<0.001, n>3.
