Determination of the optimum concentration of cisplatin and the transfection efficiency of the MTDH intervention sequence. (A) A curve was drawn to elucidate the optimum concentration of cisplatin, with cell viability on the y-axis and cisplatin concentration (0–50.0 μg/ml) on the x-axis, indicating that treatment with 1.0 μg/ml cisplatin for 24 h was appropriate for the assessment of prostate cancer cell chemotherapy sensitivity. (B) Compared with image in light microscopy, the cells transfected with MTDH siRNA present green fluorescent protein in image in fluoroscopy. The results indicate that >80% of cells were observed to be transfected, suggesting successful transfection. MTDH, metadherin; siRNA, small interfering RNA.
