The expression levels of survivin (-WT and -2B) in RA-FLS are upregulated by PDGF, but not by IL-1β, TNF-α or leptin.(a) Cultured primary synoviocytes from RA patients were analyzed for survivin-WT and -2B expressions using a Western blot analysis. The survivin expression levels in the adherent cultured primary synoviocytes decreased with the cells passages. The cropped blots were used in the figure. (cg) RA-FLS (n = 3 from different RA individuals) were cultured in the presence of (c) PDGF (10 ng/ml), (e) IL-1β (1 ng/ml), (f) TNF-α (20 ng/ml) or (g) leptin (1000 nM) for 24 to 48 hours and (d) PDGF (from 0.1 to 10.0 ng/ml) for 48 hours, after serum starvation (0.1%FBS/DMEM) for 24 hours. The time- and dose-dependent relative expression levels of survivin-WT and -2B were analyzed by real-time PCR. The relative expression levels were corrected based on the level of HPRT. Adherent FLS were used for experiments (without (a)) at passages 3 through 6. The results are the Means ± SEM (standard error of the mean). The Dunnett method, ∗∗p < 0.01, p < 0.05.
