Loss of PA-X expression increases cell death in DEF. (A) Flow cytometry analysis of cells infected with CK10 or PA-X-deficient viruses and of mock-infected control cells at 12 and 24 h p.i. Cells were stained with FITC-conjugated annexin V and propidium iodide (PI). Apoptotic cells are those with high annexin V and low PI staining, whereas necrotic cells are highly stained with both annexin V and PI. The percentages of positive cells among the total numbers of cells analyzed are shown. (B and C) Quantitative analyses of wt or PA-X-deficient virus-induced apoptosis (B) and necrosis (C) by flow cytometry. Values are proportions of the total number of cells that are apoptotic or necrotic and are means ± SD of the results from at least three independent experiments.*, P < 0.05; **, P < 0.01.
