(A, B) Cells were treated with 1 μM PARPi (24 h) and harvested for preparation of metaphase spreads. Left panels: dot plots indicating the total amount of aberrations per cell. Right panels: histograms depict PARPi-induced chromosomal aberration load relative to empty vector-transduced cells. At least 100 metaphases were scored for chromosomal aberrations for each condition. Results are mean ± SD of two (A) and four (B) independent experiments, respectively. (C, D) Cells were treated with 1 μM PARPi continuously for 10 days, after which culture dishes were stained with crystal violet and colonies containing >50 cells were counted. Results are mean ± SD of three independent experiments. For (B), (C) and (D), statistical significance was determined with two-tailed unpaired and paired Student's t-test, respectively; *P < 0.05 compared to empty vector-transduced cells.
