(A) The secreted level of VEGF was determined by ELISA assay. VEGF secretion level was negatively regulated by CHIP in both 786-O and OS-RC-2 cells, respectively. All experiments were carried out in triplicate. (B) Representative WB analysis of the relative protein levels of VEGFR2 in CHIP restoration and control group for both 786-O and OS-RC-2 cell lines. VEGFR2 protein level was dramatically reduced after CHIP restoration. (C) Model of mechanism of CHIP inhibiting RCC angiogenesis. CHIP decreased the secreted level of VEGF, meanwhile increased the degradation of VEGFR2 and downregulated the protein level of VEGFR2 on cell membrane. Data are shown as mean ± s.e.m. *P < 0.05.
