Supplemental Figure S10

Dose-dependent inhibition of human intestinal fibroblast (HIF) cytokine production by antibodies to type I IL-1 receptor (IL-1RI) and IL-1α but not IL-1β. A: A neutralizing anti-human IL-1RI (0.1 to 2.5 μg/mL) antibody was added to HIFs for 1 hour followed by epithelial necrotic cell lysates (NCLs) (diluted 1:20), and cells were incubated for 24 hours. B and C: Neutralizing anti-human antibodies to IL-1α (0 to 100 ng/mL) (B) or IL-1β (0 to 5000 ng/mL) (C) were incubated with NCLs for 1 hour, and then lysates were added to HIFs for 24 hours. The levels of IL-6 and IL-8 were measured in HIF supernatants by enzyme-linked immunosorbent assay. P < 0.05, ∗∗P < 0.01, and ∗∗∗P < 0.001. n = 3 (A and C); n = 4 (B).
