DNA, histones, and chromatin do not mediate intestinal epithelial necrotic cell lysate (NCL)–induced human intestinal fibroblast (HIF) cytokine production. HIFs were incubated in medium alone or in the presence of HT29-derived genomic DNA, soluble histones, or sheared chromatin for 24 hours; the concentrations of DNA, soluble histones, and chromatin were equivalent to those in a 1:20 dilution of the NCLs of the same cells. NCLs increased IL-6 production, but none of the three nuclear components induced HIFs to produce IL-6. IL-6 levels in the supernatants were measured by enzyme-linked immunosorbent assay. n = 4.
