Effect of troglitazone (TGZ) on SiHa cell apoptosis.

Notes: SiHa cells were treated with TGZ (400 μg/mL) for 24, 48, and 72 hours. (A) Axis labels show the apoptosis rate of SiHa cells. Flow cytometric analysis of TGZ-induced apoptosis in SiHa cells using annexin V–fluorescein isothiocyanate/propidium iodide staining. Cells in the lower right quadrant represent early apoptotic cells, and those in the upper right quadrant represent late apoptotic cells. Apoptosis rate = early apoptotic cells + late apoptotic cells. (B) The percentage of apoptotic cells is presented as the mean ± standard deviation. The data are representative of three similar experiments. The apoptosis rate for SiHa cells was 2.97%±1.29% (0 hours), 3.87%±1.0% (24 hours), 5.20%±1.14% (48 hours), and 6.10%±1.49% (72 hours) after treatment with 400 μg/mL TGZ, without significant differences at each time point (P=0.06). (C) Cell morphology was observed in each time group using an inverted optical microscope (200×). Scattered polynuclear apoptotic cells were observed. No significant differences were observed for the varying lengths of incubation.
