The mAbED-C99 promotes APP β-secretase processing in vivo. TgAPPwt female mice at 8 months of age were treated with mAbED-C99 or control IgG1 at 5 μg/mouse by intracerebroventricular (i.c.v.) injection and killed 24 h after the treatment (n=6). (a) Cytosolic- (left panels) and membrane-associated proteins (right panels) prepared from mouse brain homogenates were subjected to WB analysis for APP processing. (b) For WB quantitative analysis, band density ratio of membrane-associated β-CTF to membrane-associated total APP was analyzed (upper panel). Aβ40/42 was also analyzed by ELISA (lower panel, n=6).
