(A) Cyclin A2-specific or control siRNAs were transfected into HeLa cells for 24 h, and then the cells were treated with or without 9.2 μM HDAB for an additional 24 h. The cell cycle distribution was determined by flow cytometry, and representative images are shown (left panel). Three independent experiments were performed; the S phase data are presented as the mean ±SD (column, *p < 0.05). (B) HeLa cells were pretreated with 50 nM (F50) or 100 nM (F100) flavopiridol for 2 h and then treated with or without 9.2 μM HDAB for an additional 24 h; the cell cycle distribution was analysed by flow cytometry. The data from three independent experiments are presented as the mean ±SD (*p < 0.05). (C) Cells were treated with or without 9.2 μM HDAB in the presence or absence of the indicated concentrations of flavopiridol, and apoptosis was examined (*p < 0.05). (D) HeLa cells were treated with or without 3.7 μM HDAB in the presence or absence of 100 nM flavopiridol, and colony formation was examined (*p < 0.05).
