H3.3K36M and SETD2 deletion are reported to affect global H3K36me3 and therefore are expected to impair overall BS69 binding to chromatin. H3.3G34R/V, on the other hand, affects H3.3K36me3 only in cis and thus will alter BS69 binding only locally. The major cancer association of each mutation is indicated.
