(A, B) RBL-2H3 cells (5 × 105/well) and RPMCs (2 × 104/well) were sensitized with anti-DNP IgE (50 ng/ml). After incubating overnight, the cells were pretreated with or without drugs including tyrosol, GA, and Dexa for 1 h and then challenged with DNP-HSA (100 ng/ml). Histamine level was assayed using the o-phthaldialdehyde spectrofluorometric procedure. (C) The level of β-hexosaminidase was measured using β-hexosaminidase substrate buffer. (D) RBL-2H3 cells (3 × 104/well) were pretreated with or without tyrosol for 24 h and then incubated with 1 mg/ml MTT for 2 h. The absorbance intensity was detected using a spectrophotometer. Each data presented as a graph represents the mean ± SE of three independent experiments. *Significant difference from DNP-HSA challenged group at P < 0.05. GA: gallic acid; Dexa: dexamethasone.
