Overexpression of SD2 in neurons blocks CCG-17444 activity. P19-derived neurons were transfected with a control vector or SD2 expression vector (Shroom3 aa 1,762–1,986) and treated with DMSO (vehicle control) or 25 μM CCG-17444 for 24 h prior to analysis of neurite length. Ectopic expression of wild type but not Cys254Ala SD2 reverses the action of CCG-17444, demonstrating that the mechanism of action of CCG-17444 in neurons is inhibition of SD2 function (n = 3, *p < 0.0001, **p < 0.05 Student’s t test).
