Effect of Rad50 knockdown on AAV replication in the presence of HSV-1. (A) Western blot analysis of HeLa shRad50 (R) and HeLa shLuc (L) cells. (B and C) HeLa shRad50 or HeLa shLuc cells were coinfected with wt AAV (MOI, 10³ GP/cell) and wt HSV-1 (MOI, 5 PFU/cell). Twenty hours postinfection (p.i.), DNA was extracted, and the amounts of AAV and HSV-1 genomes were quantified by qPCR. Results are expressed as the ratio of rep or UL19 to β-globin genes (mean and SD).
