Validation of the binding mode and specificity of catechol bisphosphate derivatives. a) Structure of the BODIPY-FL-labeled catechol bisphosphate derivative 10 and principle of the assay. b) 10 nm of 10 were incubated with the indicated wild-type and mutant STAT5 proteins at the indicated protein concentrations. Binding was detected by an increase in fluorescence polarization. Kd values for the STAT5b mutants and STAT5a could not be determined, since their extrapolated values exceed the highest protein concentration tested (2560 nm).
