For each of five bones, c. 2.5 g of bone powder was homogenized and 400 mg distributed into 6 tubes labelled A–F. A digestion buffer was added to all samples at time (t) = 0 and samples were vortexed and left on rotator at 50 °C. At t = 30 minutes, the pre-digestion was removed from sample A and a new digestion buffer was added, followed by a full 24-hour incubation. Similarly, pre-digestions were removed from samples B–E at respectively 1 hour, 2 hours, 3 hours, 6 hours. No pre-digestion was removed from sample F.
