COX-2 downregulates miR-23b, miR-146b, and miR-183 in NCL cells. NCL-V and NCL-C cells were transfected with 50 nM scrambled siRNA or 50 nM siCOX-2 or were stimulated with 5 μM PGE₂. COX-2 protein levels are shown by Western blotting. After 48 h, total RNA was extracted and miRNA expression was analyzed by real-time PCR and normalized to U6 RNA expression. The expression level was determined relative to that in NCL-V cells (arbitrarily assigned a value of 1).
