Faap20−/− or wild type mice at 6-8 weeks of age were injected with one dose 8mg/kg MMC to induce the bone marrow failure. PBS injected Faap20−/− or wild type mice were used as controls. Mice were analyzed 8 days after injection. (A) The early erythroid progenitors in Faap20−/− mice were depleted after MMC exposure. Freshly dissociated bone marrow cells were labeled with CD71 and Ter119 mAb, and subjected to flow cytometric analysis. The five regions (R1 to R5) within a flow cytometry scatter plot contain distinct cell types found in the different stages of erythrocyte differentiation were selected as indicated. There are predominantly proerythroblasts in region R1, basophilic erythroblasts in region R2, late basophilic and chromatophilic erythroblasts in region R3, orthochromatophilic erythroblasts in region R4, and reticulocytes and erythrocytes in R5.
