The contractile activities of TRPC3 overexpressing fibroblasts were analyzed using a gel contraction assay. Each gel contained either 8 × 105 TRPC3 overexpressing cells or empty vector transfected control cells and was incubated for 24 hours. After starvation, the gels were treated with either a TRPC3 agonist (OAG) or a TRPC3 inhibitor (Pyr3) and the contraction ratio was observed. Gels with TRPC3 overexpressing cells exhibited more contraction than control gels. This contraction effect was enhanced with OAG and attenuated by Pyr3.
