Mixed dominance of the constitutive RGC2-7A mutant. Arsenite sensitivity of the RGC2-7A mutation. The RGC2-7A allele was integrated into the yeast genome at the RGC2 locus of a strain bearing an rgc1Δ mutation. Diploids homozygous for the rgc1Δ mutation were tested for arsenite sensitivity. Equivalent numbers of cells in 10-fold serial dilutions of each strain were spotted onto YEPD plates with or without the indicated concentration of arsenite and incubated for 3 days at 30°C. Yeast strain genotypes were RGC2/RGC2 (DL4112), RGC2/RGC2-7A (DL4113), and RGC2-7A/RGC2-7A (DL4114).
