Male or Female BDF1 mice were lethally irradiated and transferred with TCD-BM alone or plus 4 x 106/mouse CD25-depleted total T cells from normal B6 Ly5.1+ donors. HY-specific iTregs were also transplanted at 2 x 106/mouse on day 0 of BMT. Fourteen days after Teff injection, recipient spleen and liver were harvested and total T cells isolated, 4 mice per group. (A) Representative flow analysis of IFNγ and TNFα with effector CD4 and CD8 T cells. B) Represents the absolute number of CD4 and CD8 Teffs secreting IFNγ and TNFα within the recipient spleen. (C) Serum was collected on day 14 and cytokine levels were assessed by cytometric bead analysis, one representative analysis of two independent experiments.
