Effect of temperature and pH on UPF0586 protein activity. Temperature (A) and pH dependence (B) of enzymes were determined with the use of homogeneous recombinant yeast, chicken, rat, or human UPF0586 protein (yUPF0586, chUPF0586, rUPF0586, or hUPF0586, respectively). Enzyme preparations (0.10–0.35 μg of protein) were incubated for 20 min in the presence of 1 μm [1H + 3H]SAM (100 pmol, 400–600 × 103 cpm) and 20 mm carnosine, as described under “Experimental Procedures.” Values are the means ± range (error bars) of two separate experiments. When no error bar is shown, the error is smaller than the width of the line.
