Effect of EXTL2 overexpression on FGF2-induced signaling. Three clones stably overexpressing EXTL2 (L2cl.1, L2cl.2, and L2cl.3) wild-type (wt) and mock-transfected (Mock) cells were starved in serum-free medium for 18 h and then stimulated with 10 ng/ml FGF2. After 10 min, cells were lysed, and the cell lysates were subjected to SDS-PAGE followed by Western blotting using phosphospecific ERK (pERK1/2), total ERK, and GAPDH antibodies. The lower panel shows the relative intensity of immunoreactive bands of phosphorylated versus total ERK normalized to the mock-transfected cells.
