Growth of RNase III deletion mutants in liquid and on solid medium

(a) Cells suspended at an OD 730 nm = 0.05 (Tecan) were tenfold serially diluted four times in Media A+, spotted on solid medium, and grown for 65 h prior to being photographed. A representative of triplicate plates is shown. Liquid cultures of (b) single, (c) double, and (d) triple RNase III mutants were inoculated at an OD 730 nm = 0.05 (Tecan) and grown in 12 well culture plates on a shaking platform (150 rpm). Error bars represent s.d. of three separate cultures. All growth experiments were carried out in ambient CO2 at 38°C with a light intensity of 150 μmol photons m−2 s−1.
