The inflammatory cytokines IL-1β and TNF-α upregulate FHC protein levels in neurons, but only TNF-α alters FHC mRNA. A, Rat cortical Neurobasal neurons were treated with IL-1β (10 ng/ml), TNF-α (10 ng/ml), or morphine (Mor.; 1 μm) for 24 h. Both inflammatory cytokines cause a significant increase in neuronal FHC protein (n = 3; compared with vehicle). B, Neurobasal neurons were treated with either IL-1β or TNF-α (10 ng/ml) for 6 h, and total RNA was collected. FHC transcript levels were assessed via qPCR, and significant alterations are observed only for TNF-α (n = 3).
