Figure 3. — Gram-scale, CHO-based antibody production. CHO-S cells were transfected with an antibody expression plasmid (2 µg DNA/1 × 10⁶ cells) via small-scale (8 × 10⁷ cells) or large-scale (1 × 10¹⁰ cells) electroporation. Sodium butyrate (1 mM) was added to cultures and the temperature lowered to 32 °C 24 h postelectroporation. Cultures were fed daily using supplemented CD OptiCHO medium. (A) Secreted antibody titers were measured on days 6, 8, 10, and 12 following large-scale electroporation and on days 6, 8, 10, 12, 14, and 16 following small-scale electroporation. Total IgG production following (B) small-scale and (C) large-scale electroporation was calculated for days 6 to 12.
