Binding site selection for maize-TGA1 and teosinte-TGA1. α-32P-labeled DNA fragments that contain 26 randomized nucleotides were incubated with maize or teostine-TGA1. After five rounds of EMSAs, the shifted DNA bands were excised, and the consensus binding motif was obtained. Both maize-TGA1 and teosinte-TGA1 proteins bind to DNA fragments with a GTAC core motif.
