Fv1 induces cleavage of Poly ADP ribose polymerase 1 (PARP), but cell death is not blocked by the caspase inhibitor zVAD-fmk. (A) 24 h after seeding, Panc89 cells were treated with Fv1. At the indicated time points, whole cell lysates were analyzed by Western blotting for the cleavage of PARP; (B) Panc89 cells were treated with Fv1 as described above, but in combination with zVAD-fmk for 24 h. Afterwards, cells were stained with Annexin V and propidium iodide and measured by flow cytometry. Viability of Panc89 cells treated with Fv1 and zVAD-fmk was observed by AlamarBlue staining after 72 h of treatment (see Supplementary Figure S4).
