VEGF induction of MEF2C RNA and protein expression in a time- and dose-dependent manner in human endothelial cells. (A) HRECs and HUVECs were treated with VEGF at the indicated doses for 4 hours. Total RNA was isolated, and reverse transcription and quantitative real-time PCR were performed as in Figure 1. Values were normalized to GAPDH gene expression. (B) HRECs were treated with VEGF (25 ng/mL) for the indicated amount of time. Total RNA was isolated, and MEF2C expression was assessed by quantitative real-time PCR. Results are representative of three independent experiments, each performed with duplicate samples. CN, control. (C) HRECs and HUVECs were treated with VEGF (25 ng/mL) for the indicated amount of time. Cells were harvested and probed with antibody to MEF2C. The same membrane was stripped and blotted with antibody to GAPDH for normalization.
