FIGURE 4.

GP12 exhibits a preference for glycosylated WTA analogs. Activity was assayed by monitoring the disappearance of full-length [14C]glycerol-labeled WTA analogs by anion exchange HPLC. Data points are the mean from three independent experiments and error bars are the S.D. Insets show plots of the initial rates as a function of GP12 concentration and enzyme turnover, calculated as the slope of the linear regression line, was 6.2 and 80 μm min−1, for non-glycosylated lipid ϕ.40 and glycosylated lipid ϕ.40, respectively. A, reactions contained lipid ϕ.40 analog at 135 μm and GP12 at 0.08 (●), 0.16 (○), 0.32(■), and 0.64 (□) μm. B, reactions contained glycosylated lipid ϕ.40 analog at 138 μm and GP12 at 0.02 (●), 0.04 (○), 0.08 (■), and 0.16 (□) μm.
