Northern blot to validate the data from Fig. 3B in another cell line (HEK293T). The cells were transfected with the indicated combinations of shRNA and TuD constructs, and both TuD and the two shRNA strands were detected using specific probes. Note that again there was no evidence for shRNA tailing and trimming, supporting our conclusion that the mechanism of TuD-induced shRNA inhibition is likely to be stable sequestration and thus is different from the previously reported TuD action on miRNAs (see main text).
