4LB5 is internalized by target cells. MDA-MB-231 cells were incubated for 6 h at 37 °C (A) or 4 °C (B) with Cy5-labeled 4LB5. Cells were then harvested and analyzed using a FlowSight instrument (AMNIS) to acquire bright field (Ch01), Cy5 (Ch11), and merged images. At least 10.000 cells were acquired for each experimental point. (C and D) Internalization analysis was performed using the FlowSight Internalization wizard and quantification of cells internalizing 4LB5 in the two conditions is reported.
