Compound-virus interactions. (A) JUNV GP-pseudotyped lentiviral vectors were incubated with DMSO or compounds at an inhibitory concentration of 2 μM (compounds 1 and 2) or 3 μM (compound 3) for 1 h and then added to 293A cells and medium so that dilution resulted in a subinhibitory concentration of 0.05 μM (compounds 1 and 2) or 0.075 μM (compound 3). (B and C) 293A cells were incubated with DMSO or compounds 1 h before the addition of JUNV GP-pseudotyped lentiviral vectors, at the inhibitory concentration (B) or subinhibitory dose (C). All cells were assayed for GFP at 48 to 72 h. Graphs show the resulting inhibition of JUNV GP entry after treatments. Relative titers are shown as percentages of the control titer (DMSO treated). The mean titer on DMSO-treated cells was 1.6 × 107 TU/ml. All data are the means ± standard deviations of results from 2 to 3 independent experiments. Statistical significance compared to the DMSO control is indicated (***, P < 0.001; ****, P < 0.0001).
