Apparent molecular mass of rice P5C dehydrogenase under native conditions. Aliquots (100 μL) of purified recombinant rice P5C dehydrogenase, adjusted to 0.5 mg mL⁻¹, were subjected to gel permeation chromatography on a Superose 12 HR 10/30 (Pharmacia) column that had been equilibrated with 50 mM Tris-HCl buffer, pH 7.75, containing 250 mM NaCl. A molecular mass of 259 ± 14 kDa was estimated for P5C dehydrogenase, suggesting a homotetrameric composition of the enzyme. Molecular weight markers used (Pharmacia 17-0442-01 and 17-0441-01) are specified in Materials and Methods.
