Subcellular localization of GFP-HDHB is cell cycle dependent. (A) GFP-HDHB transiently expressed in U2OS cells in G1 or S phase was visualized by fluorescence microscopy. Nuclei were stained with Hoechst dye. Bar, 10 μm. (B) Subcellular localization of GFP-tagged HDHB in asynchronous, G1, and S phase U2OS cells was quantified. The number of GFP-positive cells with a given distribution pattern was expressed as a percentage of the total number of GFP-positive cells in that experiment. (C) Cytoplasmic and nuclear extracts of synchronized U2OS cells (G1 and S phase) were analyzed by denaturing gel electrophoresis and Western blotting with antibody against recombinant HDHB, α-tubulin, and PCNA. Immunoreactive proteins were detected by chemiluminescence.
