Proposed model on how STAT3 and myocardin regulate VSMC phenotypic switch stimulated by VEGF. VEGF activates STAT3 via phosphorylation. The activated STAT3 then suppresses myocardin activity, which results in reduced expression of contractile genes through an as yet undefined mechanism and increased proliferation markers in the VSMCs. Both myocardin and STAT3 can enhance the expression of VEGF, thus creating a feedback loop to regulate VSMC phenotypic switch.
