ZFP regulate the expression of TNF through its 3′UTR. The regulatory effect of 50 CCCH-containing ZFP on the expression of GFP-mTNF3′UTR- and GFP-hTNF3′UTR reporters was evaluated by cotransfection in HEK293T cells. A mCherry expression vector was used as a transfection control and an empty vector (Control) was used to measure the basal expression of the reporters. GFP mean fluorescence intensity (MFI) was measured in mCherry-positive cells by FACS, and results are expressed as relative MFI compared with control. Results represent the mean ± SEM of three independent experiments.
