Intracellular 3OC12 acid accumulation is PON2 dependent. (A and B) HEK and EA.hy cells were treated for the times shown with 25 μM 3OC12, rinsed, and lysed with acetonitrile, and lysates were analyzed for 3OC12 acid by HPLC. (C) HBEC were transfected with scrambled or PON2 siRNA for 3 days and then treated for 5 min with 25 μM 3OC12. Cells were then rinsed and lysed, and lysates were analyzed for 3OC12 acid. PON2 activity in scrambled and PON2 siRNA cell lysates was 16.8 ± 2.9 U/mg and 5.2 ± 0.9 U/mg, respectively (C). Data are the means of three separate experiments ± standard deviations.
