XN does not inhibit ATF6 processing. Huh-7 cells were cultured with medium A for 48 h and then incubated with or without 250 nm Tg for 5 h. Cells incubated in Tg were then switched to medium A in the presence of 250 nm Tg alone or Tg plus 300 μm AEBSF or 30 μm XN. After incubation for 3 h, whole cell extracts were subjected to immunoblotting (IB) with anti-ATF6, anti-SREBP-1 (2A4), or anti-β-actin antibodies. The same results were obtained in more than three separate experiments.
